Vinca major subsp. hirsuta’nın antioksidan aktivitesinin ve RP-HPLC-UV ile fenolik bileşenlerinin belirlenmesi

Bu çalışma, Vinca major subsp. hirsuta’nın fenolik içeriğinin ve antioksidan aktivitesinin belirlenmesi amacıyla yürütülmüştür. Bitkinin fenolik içeriğinin belirlenmesi için ters fazlı yüksek performanslı sıvı kromatografisi (RP-HPLC-UV) kullanılmıştır. Toplam fenolik madde, FRAP ve DPPH radikal temizleme yöntemleriyle bitkinin antioksidan aktivitesi belirlenmiştir. Bitki yaprak, çiçek ve sap olmak üzere üç kısma ayrılmıştır. Uygulanan antioksidan yöntemlere göre bitkinin yaprak kısmı en yüksek aktivite göstermiştir. RP-HPLC-UV ile Vinca major subsp. hirsuta.’ın tüm kısımlarında ve ekstraktlarında değişik konsantrasyonlardaki 5 farklı fenolik bileşen tespit edilmiştir. Klorojenik asit 5344,93 ila 112,97 µg phenolic bileşen/g kuru örnek birimle ana bileşen olarak belirlenmiştir. Üç örnekte de en düşük fenolik bileşen gallik asit olarak bulunmuştur. Bitkinin üç parçasında en yüksek fenolik bileşen içeriğini ise yaprak göstermiştir. The aim of the study is to determine phenolic composition and antioxidant activity from Vinca major subsp. hirsuta. Reverse phase high performance liquid chromatography (RP-HPLC-UV) was used to determination phenolic compounds. The antioxidant activity of the plant was investigated in terms of total phenolic content, FRAP, and DPPH radical scavenging assays. The plant is divided into three parts: leaf, flower and stem. According to the applied antioxidant assays, leaf of the plant showed the highest activity. 5 different phenolic compounds in the range of concentrations were determined in the all extracts and parts of Vinca majör subsp. hirsuta by RP-HPLC-UV. Chlorogenic acid was the main ranging from 5344.93 to 112.97 µg phenolic compound/g dry sample, respectively. Gallic acid was found to be lowest in the three parts of the samples. The leaf was observed highest phenolic compounds between the three samples.
