Mazı meşesi (Quercus infectoria) bitkisinin antioksidan ve antimikrobiyal aktivitesi ile uçucu yağ ve fenolik bileşenlerinin belirlenmesi

Özet
Mazı meşesi (Quercus infectoria) kozalağının toplam polifenol ve toplam flavonoid madde miktarları ölçülmüş aynı zamanda antioksidan kapasitesi; FRAP, CUPRAC ve DPPH? radikali temizleme yöntemleriyle açığa çıkarılmıştır. Elde edilen verilere göre toplam polifenol ve flavonoid madde içerikleri 342.87±20.34 mgGAE/g numune, 0.75±0.07 mg kuersetin/g numune şeklindedir. Antioksidan özellikleri ise FRAP için, 4.04±0.21 ?mol FeSO4.7H2O/g numune, CUPRAC için 13.23±0.51 mmol TEAC/g numune olarak belirlenmiştir. DPPH? için ise IC50=0.002 mg/mL değeriyle çok iyi antioksidan özellik"sergilediği hatta o alan için standart olarak kullanılan troloksa çok yakın değerde aktivite gösterdiği görülmüştür." Antimikrobiyal aktivite için disk difüzyon ve minimum inhibisyon konsantrasyon (MİK) yönteminden yararlanılmıştır. Mazı meşesi kozalağının kullanılan test mikroorganizmalarına karşı 250-4000 µg/mL değeriyle antibakteriyel aktivite gösterdiği, maya suşlarına karşı ise 125 veya 250 µg/mL değeriyle, özellikle de Candida glabrata ATCC 2001 ve Candida krusei ATCC 6258'e karşı 125 µg/mL değeriyle antifungal özellik gösterdiği bulunmuştur. Mazı meşesi'nden açığa çıkarılan uçucu yağın SPME ve GC/MS teknikleriyle aydınlatılması sonucunda 29 tane bileşen tespit edilmiştir. (Z)-Anethole eteri (%28.55), pentadecanolide esteri (%26.44), diethyl phthalate esteri (%6.46) ve acetoin ketonu (%5.66) bileşikleri en yüksek oranlara sahip oldukları için ana bileşenler olarak tanımlanmıştır. Mazı meşesi'nin fenolik bileşen tayini için HPLC-UV yönteminden faydalanılmış ve analizde 15 adet standart kullanılmıştır. Mazı meşesi kozalağında sadece gallik asit %2.9 (20.56 ppm) fenolik bileşiğine rastlanmıştır.
