dc.description.abstractBu çalışma interlökin-6 (IL-6) geni 572 G/C polimorfizminin frekanslarını belirlemek ve tip-2 diyabet, diyabetik nefropati ve diyabetik retinopati gelişiminde bu polimorfizmin rolünü incelemek için yapılmıştır. Bu çalışmada, 210 genomik DNA örneği analiz edildi. Genomik DNA, DNA izalosyon kiti kullanılarak periferal kandan elde edildi. IL-6 genindeki 572 G/C polimorfîzmi, PCR, restriksiyon fragment uzunluk polimorfizmi (RFLP) ve elektroforez yöntemleri kullanılarak belirlendi. IL-6 geninin PCR ürünleri BsrBI restriksiyon enzimi ile muamele edilerek kesildi ve %2'lik agaroz jel elektroforezde incelendi. Sonuçlar istatistiksel olarak değerlendirildi. Buna göre; bu çalışmada, G572C genotip frekansı kontrol grubu için GG %83.1 GC %16.9 ve CC %0.0 olarak, hastalarda için GG %88.2, GC %9.4 ve CC %2.4 olarak tespit edildi. Genotip frekansı açısından kontrol ve hasta grubu arasında istatistiksel olarak önemli fark bulunmadı. Kontrol grubunda ise hasta grubuna göre GC genotipinin önemli düzeyde yüksek olduğu görüldü. Sonuç olarak, IL-6 geni 572 G/C polimorfizmi ile tip-2 diyabet hastalığı ve diyabetik nefropati ve diyabetik retinopatinin ilişkili olmadığını söyleyebiliriz.
dc.description.abstractThis study was performed on determine the polymorphism frequencies of the ınterleukın-6 (IL-6) gene 572G/C and to examine the role of this polymorphism in type-2 diabetes, diabetic nephropathy and diabetic retinopathy development. Genomic DNA obtained from 210 persons was used in the study. IL-6 gene 572G/C polymorphism were determined using the PCR, restriction fragment length polymorphism (RFLP) and electrophoresis. IL-6 gene PCR products was discontinued by treatment with restriction enzyme BsrBI, and were analyzed in 2% agarose gel electrophoresis. The results were statistically analyzed. Accordingly; In this study, The frequencies of G572C genotypes, in controls GG 83.1%, GC 16.9%, CC 0% and in Type-2 patients GG 88.2%, GC 9.4%, CC 2.4% were found. There was no statistically significant difference between the control group and the patient groups in genotype frequencies. It was determined that GC genotype frequency increases significantly in control according to patient group. As a result, we can not say that there is an interaction between IL-6 gene 572 G/C polymorphism with type-2 diabetes, diabetic nephropathy and diabetic retinopathy.
