    The effect of bee pollen on reproductive and biochemical parameters in methotrexate-induced testicular damage in adult rats

    Methotrexate (MTX) is an anticancer drug used in chemotherapy. MTX was known for its toxic effects involving most of the organs including testis. Bee pollen is healthy food for human and has antioxidant effect. We intended to determine protective effect of bee pollen against testicular injury caused by MTX in rats. Thirty-two adult Sprague Dawley male rats were used, and 4 groups were formed: control, MTX, pollen, and MTX + pollen. Rats were given pollen at a dose of 400 mg/kg with intragastric gavage for 10 days. On day 7, MTX was administered a single dose of 30 mg/kg ip. Serum testosterone and LH, tissue MDA level, and SOD and CAT enzyme activities were examined. In addition, spermatological parameters were evaluated. MDA level and SOD activity increased while testosterone level decreased significantly in the MTX group compared to the control group. In the MTX + pollen group, MDA level and SOD activity decreased while testosterone level increased. There was no significant change in CAT activity and LH values. Abnormal sperm ratio decreased in the MTX + pollen group compared to the MTX group. Our results suggest that bee pollen has a healing effect on reproductive parameters in testicular damage caused by MTX.

    The effect of hesperidin and quercetin on oxidative stress, NF-kappa B and SIRT1 levels in a STZ-induced experimental diabetes model

    Objective: The aim of this study is to investigate the roles of SIRT1 and NF-kB in the pathogenesis of diabetes mellitus in rats with STZ-induced diabetes and determine the effects of hesperidin and quercetin on oxidative stress and on the levels of SIRT1 and NF-kB. Materials and methods: The experimental animals were divided into four groups, each group comprising ten rats designated as follows: group 1 served as control rats (C); group 2 served as diabetic rats (DM); group 3 served as diabetic rats administered hesperidin (DM + HSP) (100 mg/kg b.w.) in aqueous suspension orally for 15 days; and group 4 served as diabetic rats administered quercetin (DM + Q) (100 mg/kg b.w.) in aqueous suspension orally for 15 days. Results: In diabetic group, liver and kidney SIRT1, SOD and CAT activities were significantly lower than control group (p < 0.05). Hesperidin and quercetin caused significant increase in the SIRT1, SOD and CAT activities of both DM + HP and DM + Q groups kidney tissues compared to DM group (p < 0.05). Liver SOD activies were not found to differ significantly between DM, DM + Q and DM + HP groups (p > 0.05). In DM + HP group, liver CAT activities were significantly higher than DM (p < 0.05), but there was no significant difference in liver CAT activities between DM and DM + Q (p > 0.05). In diabetic group, liver and kidney NF-kB and MDA levels were increased compared to control group (p < 0.05), and groups of DM + HP and DM + Q had lower NF-kB and MDA levels than diabetic group (p < 0.05). Conclusion: As a conclusion, based on the results we obtained from this study and the literature data discussed above, we determined in STZ-induced diabetic rats that, increased glucose levels and liver and kidney damage markers decreased significantly after administration of hesperedin and quercetin, and that oxidative stress and NF-kB levels increased while SIRT1 levels decreased in the diabetic group

    Effect of white tea consumption on serum leptin, TNF-α and UCP1 gene expression in ovariectomized rats

    Background: Obesity and dyslipidemia due to estrogen deficiency are among the important health problems in menopausal women. Increasing evidence reports the anti-obesity and anti-hyperlipidemic properties of tea polyphenols. However, the effect of white tea (WT) with high polyphenol content on overweight and lipid profile is uncertain. Here, we aimed to examine the effects of long-term WT consumption on serum leptin, tumor necrosis factor- alpha (TNF-?) and uncoupling protein 1 (UCP1) mRNA gene expression in ovariectomized (OVX) rats. Methods: Adult rats were divided into four groups (n = 8): (i) sham, (ii) OVX, (iii) WT and (iv) OVX + WT. WT was given at a dose of 0.5% w/v for 12 weeks. In the study, body weight, serum leptin, TNF, estradiol (E2) levels, lipid profile and UCP1 mRNA gene expression in brown adipose tissue (BAT) were evaluated. Results: There was a significant increase in body weight of OVX rats, which was decreased following WT consumption. While leptin and E2 levels decreased in the OVX group, TNF levels increased. There was no difference between the NF-?B levels of the groups. In addition, BAT UCP1 mRNA expression was significantly decreased in OVX groups, while WT treatment stimulated UCP1 activity. Conclusion: We explain the stimulatory effect of WT on weight loss mainly by the induction of UCP1 gene-mediated thermogenesis and suppression of inflammation. Therefore, we suggest that prolonged WT consumption may have beneficial effects in limiting excess weight gain caused by estrogen deficiency.

    The effect of white tea on serum TNF-α/NF-κB and immunohistochemical parameters in cisplatin-related renal dysfunction in female rats

    Objective: Nephrotoxicity is the most important side effect of the antineoplastic drug cisplatin, thereby restricting its use. The aim of this study was to investigate the protective effects of white tea infusions (WT) against renal damage induced by cisplatin (CP) in rats by biochemical and histopathological means. Materials and methods: This study used 24 female Sprague Dawley rats at 12–14 weeks of age and weighing 250–300 g. Rats were divided into three groups: Control, CP and CP + WT groups. CP was injected 7 mg/kg i.p as a single dose/rat in the CP group. White tea was given at a dose of 0.5% (w/v) for 4 weeks. At the end of the experiment, blood urea nitrogen (BUN), creatinine, uric acid, tumor necrosis factor-alpha (TNF-?), interleukin-6 (IL-6) and nuclear factor kappa B (NF-?B) along with caspase-3 in the kidney were evaluated in study. Results: BUN, creatinine, TNF-? NF-?B and IL-6 levels of the CP group showed a statisically significant increase in comparison to the control group. TNF-? NF-?B and IL-6 levels showed a statistically significant decrease in the CP + WT group with respect to the CP group. Caspase-3 levels in tubular epithelial cells decreased in CP + WT group compared with CP group (p = 0.02). Conclusion: White tea infusions reduced significantly the nephrotoxicity of CP. The anti-nephrotoxic feature of the infusion may be attributed primarily to its anti-inflammatory and anti-apoptotic characteristics.

    Effects of astaxanthin on biochemical and histopathological parameters related to oxidative stress on testes of rats on high fructose regime

    Astaxanthin (ASX) is a xanthophyll family of hydroxycarotenoids which contains sev-eral double bonds. It is produced by Haemococcus pluvialis, a microalgae and pos-sesses antioxidant and anti-inflammatory properties. The aim of this study was to test whether ASX could protect against oxidative damage in the testicular tissues of rats receiving high fructose. The rats (n = 24) were randomly divided into two main groups: control and fructose (30%, via drinking water) and then each main group ei-ther not supplemented or supplemented with ASX (1 mg kg?1 day?1, within 0.2 ml olive oil) via oral gavage. Data were subjected to two-way ANOVA. High fructose consumption tended to increase testis weight and serum testosterone concentration and decreased testicular tissue glutathione-S- transferase (GST) and superoxide dis-mutase (SOD) levels, but did not affect testicular tissue malondialdehyde (MDA) con-centration. Astaxanthin administration increased testosterone, GST and SOD levels and testis weight and decreased MDA concentration. However, ASX administration did not reverse alterations in antioxidant parameters caused by high fructose con-sumption. Inducible nitric oxide synthase (iNOS) tended to increase in sertoli cell, spermatid and spermatogonia, but not in spermatocytes and leydig cell in response to high fructose consumption. Astaxanthin administration tended to reverse elevation in iNOS in testis cells. In conclusion, ASX could help alleviate oxidative damage caused by high fructose consumption

    The effects of hesperidin and quercetin on serum tumor necrosis factor-alpha and interleukin-6 levels in streptozotocin-induced diabetes model

    Background: Diabetes mellitus (DM) is a metabolic disorder that occurs as a result of absolute or relative insufficiency of insulin release and/or insulin effect due to impairment of carbohydrate, fat and protein metabolism, and it is characterized by hyperglycemia and leads to various complications. Objective: In this study, it was aimed to investigate the effects of hesperidin (HP) and quercetin, which are natural flavonoids, on serum malondialdehyde (MDA), glutathione (GSH), tumor necrosis factor alpha (TNF??), and interleukin?6 (IL?6) levels in rats with streptozotocin (STZ)?induced diabetes. Materials and Methods: The experimental animals were divided into four groups, each group comprising ten rats designated as follows: Group 1 served as control rats (C); Group 2 served as diabetic rats (DM); Group 3 served as diabetic rats administered HP (DM + HP) (100 mg/kg b. w.); and Group 4 served as diabetic rats administered quercetin (DM + Q) (100 mg/kg b. w.). Results: Serum MDA and GSH levels were significantly higher in STZ?induced DM group than control group (P < 0.05). In DM + HP and DM + Q groups, MDA levels were significantly decreased compared to DM groups (P < 0.05), but there was no significant difference GSH levels between DM, DM + HP, and DM + Q groups (P > 0.05). TNF?? levels in STZ?induced DM group were significantly decreased compared to control group (P < 0.05), and groups of DM + HP and DM + Q had higher serum TNF?? levels than STZ?induced DM group (P < 0.05). In STZ?induced DM group, serum IL?6 levels were decreased compared to control group (P < 0.05). Conclusion: As a result, in this study, we determined that HP and quercetin may play an effective role in regulating insulin metabolism metabolism in diabetes. However, considering the incompatibility of various results in the literature as well as our own results, we think that the actual role of cytokines in the pathogenesis of diabetes is one of the issues that need to be clarified in further studies.

    Protective role of Diospyros lotus on cisplatin-induced changes in sperm characteristics, testicular damage and oxidative stress in rats

    The aim of this study was to investigate the protective effect of Diospyros lotus (DL) on cisplatin (CP)-induced testicular damage in male rats. Twenty-eight male rats were randomly divided into four groups: group 1 - control, given isotonic saline solution; group 2 - CP 7mgkg(-1) given intraperitoneally as single dose; group 3 - DL 1000mgkg(-1) per day given orally for 10days; group 4 - CP and DL given together at the same doses. CP caused a significant increase in thiobarbituric acid-reactive substances (TBARS) level and a significant decrease in superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and glutathione (GSH) levels in rats testis tissues compared to the control group. CP caused a significant increase in lipid peroxidation in testis tissues compared to the control group, whereas DL led to a significant increase in SOD and GSH levels. However, there were no statistically significant changes in GPx and CAT levels. In addition, serum testosterone levels, sperm concentration and sperm motility were significantly decreased, but abnormal sperm rate and histological changes were increased with CP. However, these effects of CP on sperm parameters, histological changes and the tissue weights were eliminated by DL treatment. In conclusion, our study showed that the reproductive toxicity caused by CP may be prevented by DL treatment.

    The effect of sulforaphane on the levels of serum cystatin-c in acetaminopheninduced nephrotoxicity in rats

    Objective: The exposure of living creatures to drugs and chemicals often results in toxicity of liver and kidney. Drugs constitute an important and big part of the community and hospital-acquired kidney diseases. In this study, we investigated the effect of sulforaphane (SFN) on the levels of cystatin-C and lipid peroxidation on acetaminophen (APAP)- induced nephrotoxicity in rats. Methods: Thirty-six Sprague-Dawley rats were separated equally into four experimental groups: control group, SFN group, APAP group, and APAP + SFN group. In the experimental treatment groups APAP was administered oral gavage at 1 g/kg 3 h after SFN treatment in last day and, in the APAP + SFN group, SFN was administered oral gavage at a dose of 500 ?g/kg exactly for three days. Rats were euthanized and sacrificed 24 h after APAP administration. Results: APAP administration showed to significant increase in serum BUN, creatinine, urea and LDH concentrations as compared to the control datas indicating the induction of severe nephrotoxicity (p<0.001). SFN treatment significantly decreased the cystatin-C levels and lipid peroxidation compared to APAP group (p<0.05). Conclusion: The present study demonstrate that the attachment of SFN to the nephrotoxicity treatment protocol will be beneficial and further studies should be conducted for cystatin C which plays an important role in kidney toxicity and disease to be routinized as a biomarker.

    Towards a comprehensive tobacco-cessation approach: A pilot-training using simulation based-learning among medical students in Türkiye

    Tobacco control (TC) is underpinned in two main strategies: preventing youth initiation and promoting tobacco cessation among users1 . Healthcare professionals (HCPs), and specially physicians, should lead TC advocacy efforts, emphasize their role as TC leaders and exemplars, and disseminate TC and tobacco-cessation training, both at undergraduate and postgraduate level. However, most physicians are neither aware nor satisfactorily competent in TC roles2 . Tobacco-cessation brief interventions can increase quit rates by 8%3 , but their implementation in clinical practice remains poor4 . In Türkiye, tobacco-cessation support is mainly provided by pulmonologists5 ; a survey reports that 41% of outpatients were asked about tobacco use, 31.8% were advised to quit, and 6% were referred to tobacco-cessation services6 . Another cross-country European survey reports that less than 50% of smokers receive cessation advice or any support to quit; cessation support is scarce and inappropriate7 . To scale-up a comprehensive tobacco-cessation approach, brief intervention should be embedded in health systems and systematically provided by all HCPs4,8. Medical School undergraduate training on tobacco cessation is crucial to shape attitudes and engage new generations of physicians2 . However, both students and educators state that the medical curriculum is insufficient regarding tobacco-cessation9,10. The international researchers we inspired and their perspectives on tobacco cessation and cessation education were influential in the realization of this project7 .
